hi and welcome to the forum. im having a little trouble with your description and a pic would help so i take it that you have an impression in some clay...?? if so mix up some plaster or resin and pour it into the impression. when set you should have a object resembling the original which you took the impression from. if its plaster or resin you can trim it up before using it ! without a pic i cant advise any further. once you have a solid object you can use it to cast from using traditional sand molding practises.

you can use any 2 part resin....car body filler...clear type of epoxy glue ...fiberglass resin... builders putty..really anything that sets hard. but some resins may set too hard to finish shaping if your impression isnt the best.

im not sure about positive or negative. i call them male and female for identification purposes. in your example your clay impression is female... after you remove your item from the clay impression you will have made a male. so you will use this male object in your sand. when its removed you will have a female mold which you pour your metal into!

I think the clay pushed onto the screw head would be a negative of the screw and a positive of the tool. Depending on your casting method you could pour plaster around the positive of the tool making a negative of the tool head.
